چکیده

The RNA-binding protein YBX1 (Y-box binding protein-1) is an epidermal stem cell function regulator. Loss of expression in keratinocytes results a decrease the proliferative potential progenitors vitro and alterations all lineages vivo. binds 3’UTR multiple targets cells, inhibits translation potent senescence-associated cytokines guarding cells against occurrence senescence, mediates cells' self-renewal proliferation. Previous studies have shown that phosphorylation can diminish its RNA affinity. Here we show phosphorylated isolated from donors different ages elevated. Small molecule inhibitors PI3 kinase (PI3K), such as PI-103 GDC-0941 inhibit levels consequently reduce cellular senescence primary adult donors. Furthermore, PI3K increase speed wound closure healing model regeneration utilizing 3D skin equivalents. Our findings uncovered new level posttranscriptional control aging by YBX1, which required to maintain regenerative potential.

Effects of Antiproliferative Protein (APP) on Modulation of Cytosolic Protein Phosphorylation of Prostatic Carcinoma Cell Line LNCaP

Antiproliferative protein (APP) isolated from conditioned media of two androgen-independent prostatic carcinoma cell lines, PC3 and Du-145 was shown to inhibit selectively cell proliferation of androgen-dependent prostate cancer cell line LNCaP in a dose dependent manner. This protein was further purified with HPLC using hydrophobic interaction column (phenyl 5PW) and was used to study the modu...

Age-associated inflammation inhibits epidermal stem cell function.

Altered stem cell homeostasis is linked to organismal aging. However, the mechanisms involved remain poorly understood. Here we report novel alterations in hair follicle stem cells during skin aging, including increased numbers, decreased function, and an inability to tolerate stress. Performing high-throughput RNA sequencing on aging stem cells, cytokine arrays, and functional assays, we ident...

Cell Elasticity Determines Macrophage Function

Macrophages serve to maintain organ homeostasis in response to challenges from injury, inflammation, malignancy, particulate exposure, or infection. Until now, receptor ligation has been understood as being the central mechanism that regulates macrophage function. Using macrophages of different origins and species, we report that macrophage elasticity is a major determinant of innate macrophage...

Epidermal stem cell diversity and quiescence

Mammalian epidermis is maintained by self-renewal of stem cells and terminal differentiation of their progeny. New data reveal a diversity amongst stem cells that was previously unrecognized. Different stem cell populations have different locations and differ in whether they are quiescent or actively cycling. During normal epidermal homeostasis, each stem cell population feeds a restricted numb...

The multifaceted adult epidermal stem cell.

Adult epidermal stem cells renew the epithelial compartment of the skin throughout life and are the most accessible of all adult stem cells. Most importantly, epidermal stem cells can be efficiently cultivated and transplanted, a significant advantage for cell and gene therapy. Recent work has pointed to the hair follicle as the main repository of multipotent stem cells in skin. Hair follicles,...
